Background Information:

UbcH7 is an E2 ubiquitin-conjugating enzyme that partners with numerous E3 ligases to catalyze ubiquitin transfer to substrate proteins. Unlike many E2s, UbcH7 preferentially works with HECT and RBR E3 ligases, including enzymes such as Parkin, RNF31 and ARIH1. UbcH7 supports both monoubiquitination and polyubiquitination, influencing protein stability, signalling, and stress responses. It is essential for the ubiquitination activity of E3 ligases such as Parkin—a major genetic factor in Parkinson’s disease. Modulating UbcH7–E3 interactions may help correct defective ubiquitination underlying neurodegeneration, inflammation, and metabolic diseases.
