Background Information:

The SCF beta-TrCP1 complex is a multi-subunit E3 ubiquitin ligase belonging to the SCF (Skp1–Cullin–F-box) family. It consists of Skp1, Cul1, Rbx1 and the F-box substrate-recognition subunit beta-TrCP1. The complex serves as a critical regulator of multiple signaling pathways, including NF-kappaB, Wnt/beta-catenin, and DNA damage response networks. Through these roles, it influences cell proliferation, differentiation, immune activation, and oncogenesis. Beta-TrCP1 can act either as a tumor promoter (e.g. through degradation of I?B-alpha and activation of NF-kappa-B signaling) or as a tumor suppressor (e.g., by targeting beta-catenin or CDC25A). This context dependence makes it a nuanced target in cancer therapeutics.
