Background Information:
The PathHunter® Human XBP1 Nuclear Translocation Cell-Based Assay Kit (U2OS) utilizes the U2OS XBP1 Nuclear Translocation cell line. The stable engineered cell line is for use in studying drug candidates affecting the Endoplasmic Reticulum (ER) stress response. This cell line enables assessment of endoplasmic reticulum stressors (e.g., Thapsigargin) via detection of resulting XBP1 nuclear translocation. This product uses EFC technology. The included cell line overexpresses PL-tagged XBP1 and nuclear localized EA. Upon pathway activation, induced nuclear translocation of XBP1 allows for complementation of these two tags and leads to production of the EFC signal.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay.
