Background Information:
The PathHunter® Human IFN gamma Signaling Reporter Cell-Based Assay Kit (HEK293) utilizes the HEK293 IFNγ Signaling Pathway Reporter cell line. This engineered cell line monitors the activation of specific signaling pathways, utilizing the established PathHunter Enzyme Fragment Complementation (EFC) technology. It can be used for studying drug candidates (small molecules or biologics) affecting the IFNγ pathway. This cell line enables assessment of IFNγ pathway stimulation based on changes in the expression of a GAS response element-driven reporter protein. The included cell line expresses PL-tagged reporter protein under the control of an IRS1 GAS-inducible transcriptional response element. Pathway activation induces expression of the PL-tagged protein, allowing detection of the EFC signal upon addition of the EA-containing detection reagent.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay.
