Background Information:

The PathHunter Human AM1 (CALCRL-RAMP2) β-Arrestin Stable Cell Line Assay (CHO-K1) utilizes the AM1 (CALCRL-RAMP2) β-Arrestin cell line. This engineered stable cell line is optimized for GPCR pharmacology and mechanism-of-action (MOA) studies, enabling quantitative assessment of β-arrestin recruitment following receptor activation for GPCR drug discovery and development programs. This cell line enables the assessment of ligand (e.g., adrenomedullin) based activation of AM1 (CALCRL-RAMP2) GPCR activity via detection of β-arrestin recruitment. This cell line utilizes Enzyme Fragment Complementation (EFC) technology, which combines an overexpressed PK-tagged CALCRL target receptor, an untagged RAMP2 protein, and an EA-tagged β-Arrestin 2 to generate a chemiluminescent signal proportional to receptor activation. This product leverages a sensitive chemiluminescent detection system that delivers a wide dynamic range and robust Z’ for potency, efficacy, and rank‑order profiling across small molecules and biologics.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay. Assay plates and positive control for use in the assays can be purchased separately. For continued use of the cell line, additional quantities of cell culture and assay reagents are available for purchase separately.
