Background Information:

The PathHunter Canine GPR91 β-Arrestin Stable Cell Pool Assay (CHO-K1) utilizes the Canine GPR91 β-Arrestin cell pool. This engineered stable cell pool is optimized for GPCR pharmacology and mechanism-of-action (MOA) studies, enabling quantitative assessment of β-arrestin recruitment following receptor activation for GPCR drug discovery and development programs. This cell pool enables the assessment of ligand (e.g., succinate) based activation of canine GPR91 GPCR activity via detection of β-arrestin recruitment. This cell pool employs Enzyme Fragment Complementation (EFC) technology that combines an overexpressed PK-tagged canine GPR91 target receptor and an EA-tagged β-Arrestin 2 to generate a chemiluminescent signal proportional to receptor activation. This product leverages a sensitive chemiluminescent detection system that delivers a wide dynamic range and robust Z’ for potency, efficacy, and rank‑order profiling across small molecules and biologics.
This kit includes a stable engineered cell pool (supplied as two vials of cryopreserved cells); cell pool specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay. Assay plates and positive control for use in the assays can be purchased separately. For continued use of the cell pool, additional quantities of cell culture and assay reagents are available for purchase separately.
