The Mas-related G protein–coupled receptor X1 (MRGPRX1) is a class A G protein–coupled receptor (GPCR) that plays an important role in sensory neuron signaling, particularly in nociception and pruritus. MRGPRX1 is activated by a range of endogenous peptides, including BAM peptides derived from proenkephalin, as well as exogenous ligands. Upon ligand binding, MRGPRX1 primarily couples to Gq and Gi proteins, leading to activation of phospholipase C, intracellular calcium mobilization, and downstream signaling pathways associated with neuronal excitation. This signaling contributes to the transmission of pain and itch sensations and to neurogenic inflammation. Through these mechanisms, MRGPRX1 participates in the modulation of peripheral sensory responses and inflammatory processes. The receptor is predominantly expressed in subsets of dorsal root ganglion neurons, where it functions as a key mediator of sensory perception.
