Background Information:

The InCELL Hunter Human TAF1L(2) Bromodomain Stable Cell Line Target Engagement Assay (HEK 293) utilizes the TAF1L(2) Bromodomain Target Engagement cell line. The stable engineered cell line is designed for use in studying small molecule drug candidates that engage with regulators of epigenetics pathways. This cell line enables assessment of compound-engagement-based stabilization of TAF1L(2) via detection of TAF1L(2) levels. This product uses EFC technology. The cell line included overexpresses TAF1L's second bromodomain fused to PL. In the absence of compound that binds and stabilizes the target, this protein fragment is naturally degraded and no EFC signal is produced. When the compound engages and stabilizes the target, EA contained in the detection reagent produces EFC signal.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay. Assay plates and positive control for use in the assays can be purchased separately. For continued use of the cell line, additional quantities of cell culture and assay reagents are available for purchase separately.
