Background Information:
Smurf1 is a HECT-type ubiquitin ligase that regulates multiple biological networks including TGFB1 (transforming growth factor beta 1) and BMP signalling pathways, the Toll-like receptor pathway, and the mitogen-activated protein kinase pathway. Smurf1 overexpression promotes tumorigenesis across multiple cancers by driving ubiquitin-mediated degradation of tumor suppressors and activating oncogenic signalling. In cervical and ovarian cancers, Smurf1 targets RhoB and ARHGAP26, respectively, enhancing invasion and metastasis via beta-catenin signaling. In breast cancer, Smurf1 facilitates estrogen receptor-alpha signaling required for estrogen-dependent tumor growth. Elevated Smurf1 expression is also linked to brain tumor malignancy, correlating with glioma grade, while its knockdown has been shown to suppress glioblastoma progression, partly through effects on PTEN. Collectively, Smurf1’s oncogenic activity is closely associated with activation of the PI3K/AKT pathway, and its suppression inhibits cancer progression in multiple tumor types.
