Background Information:
The PathHunter® Rat OPRM1 (Opioid mu) β-Arrestin Cell-Based Assay Kit (CHO-K1) utilizes the Rat OPRM1 (Opioid mu) cell line. This engineered stable cell line is optimized for GPCR pharmacology and mechanism-of-action (MOA) studies, enabling quantitative assessment of β-arrestin recruitment following receptor activation for GPCR drug discovery and development programs. This cell line enables the assessment of ligand (e.g., DADLE) based activation of rat OPRM1 (Opioid mu) GPCR activity via detection of β-arrestin recruitment. This cell line employs Enzyme Fragment Complementation (EFC) technology that combines an overexpressed PK-tagged rat OPRM1 (Opioid mu) target receptor and an EA-tagged β-Arrestin 2 to generate a chemiluminescent signal proportional to receptor activation. The assay measures ligand-induced activation by detecting β-arrestin recruitment. This product leverages a sensitive chemiluminescent detection system that delivers a wide dynamic range and robust Z’ for potency, efficacy, and rank‑order profiling across small molecules and biologics. This stable engineered cell line is supplied as two vials of cryopreserved cells. The kit includes cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating, in addition to the 10‑plate detection kit required to run the assay. All culture and assay reagents and assay detection kits are available separately to support extended studies of the cell line. Eurofins DiscoverX optimized Detection Reagents are required for optimal performance, and their use is required in working with the cell lines as part of the cell line license that you are agreeing to with the purchase of this cell line assay kit.
