Background Information:

The PathHunter Human IL-11 (IL11RA) Signaling STAT3 Reporter Stable Cell Line Assay (HepG2) utilizes the HepG2 IL11R STAT3 Signaling Pathway Reporter cell line. This engineered cell line monitors the activation of specific signaling pathways, utilizing the established PathHunter Enzyme Fragment Complementation (EFC) technology. It can be used for studying drug candidates (small molecules or biologics) affecting the STAT3 pathway. This cell line enables assessment of STAT3 pathway stimulation based on changes in the expression of a STAT3 response element-driven reporter protein. The included cell line overexpresses IL11R, and expresses PL-tagged reporter protein under the control of a STAT3-inducible transcriptional response element. Pathway activation induces expression of the PL-tagged protein, allowing detection of the EFC signal upon addition of the EA-containing detection reagent.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay. Assay plates and positive control for use in the assays can be purchased separately. For continued use of the cell line, additional quantities of cell culture and assay reagents are available for purchase separately.
