Background Information:
The cAMP Hunter™ Human DRD4 Gi Cell-Based Assay Kit (CHO-K1) utilizes the Human DRD4 cell line. This engineered stable cell line is optimized for GPCR pharmacology and mechanism-of-action (MOA) studies, enabling quantitative assessment of cAMP signaling following receptor activation for drug discovery and development programs. This cell line enables the assessment of ligand (e.g., PACAP) based activation of ADCYAP1R1 GPCR activity via detection of increasing levels of intracellular camp. This assay utilizes EFC technology to measure ligand-induced activation by quantifying intracellular cAMP levels. The cells overexpress wild type DRD4, a naturally Gi/Go coupled GPCR. Upon activation of DRD4, the decrease in levels of intracellular cAMP leads to a decreased signal from EFC when the cAMP detection kit is applied. This product leverages a sensitive chemiluminescent cAMP detection system that delivers a wide dynamic range and robust Z’ for potency, efficacy, and rank‑order profiling across small molecules and biologics. This stable engineered cell line is supplied as two vials of cryopreserved cells. The kit includes cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating, in addition to the 10‑plate detection kit required to run the assay. All culture and assay reagents and assay detection kits are available separately to support extended studies of the cell line.
